Study of UV-radiation-induced DNA repair in mouse spleen cells after whole-body 60Co-γ-irradiation

Description

A study was made of the UV-radiation-induced DNA repair, in the presence of hydroxyurea, in mouse spleen cells isolated after whole-body 60Co-γ-irradiation. In order to separate native DNA from that containing singlestranded breaks BDN-cellulose technique was applied. It was established that γ-radiation affects the UV-radiation-induced repair synthesis in mouse spleen cells. Not long after γ-irradiation (6h, 24h and 3 days), the UV-radiation-induced incorporation of 3H-thymidine in DNA, in the presence of hydroxyurea, is decreased as compared to the control. However, at later times (after a fortnight), the UV-induced unscheduled DNA synthesis was 50% as high as that in the control. After three weeks, the UV-induced DNA repair synthesis in exposed cells was the same as in the controls
