Biological evaluation of 99mTc-glucose-6-phosphate in Sprague Dawley Stock white rats (Rattus norvegicus)

Cancer is the leading cause of death by the number of patients which is predicted to increase seven-fold in 2030. Control of disease by early detection and more accurate diagnosis through the application of nuclear techniques are expected to help cure cancer at early stage. 99mTc-glucose-6-phosphate is a radiopharmaceutical that can be used for the diagnosis of cancer, and is expected to be applied primarily in hospitals that do not have the facility of PET (Positron Emission Tomography) and cyclotron. Further research has been conducted to analyse the cancer cells bioaffinity 99mTc-glucose-6-phosphate through a series of tests in animal models which has artificial cancer, among others, biodistribution tests, imaging tests, blood test clearance, and renal clearance test. Biodistribution test 99mTc-glucose-6-phosphate showed a radiopharmaceutical accumulation in the target tissue is cancerous tissue by 6.23 % at 15-minute intervals after injection. However, in addition to the cancerous tissue, this radiopharmaceutical accumulated quite high in bone that is equal to 23.99 % at 15 minutes after injection, so it will affect the imaging tests. Imaging test results showed that 99mTc-glucose-6-phosphate accumulates in the tumor/cancer tissue. It can be said that this radiopharmaceutical can be rapidly removed from the body by the results of blood clearance test and renal clearance test which indicates that the radiopharmaceutical activity in the body has dropped dramatically at 15 minutes after injection (1.25 %). Based on the results of this study concluded that the radiopharmaceutical 99mTc-glucose-6-phosphate accumulates in cancer tissue artificially. However, because there is a significant accumulation in the bone, it is necessary to reformulate this radiopharmaceutical, without using Na-pyrophosphate. (author)
