Gamma irradiation of uridine and uridylic acid in aerated aqueous solutions: identification of the major product

Description

The major product of uridine gamma irradiation in aerated aqueous solution (10-3M, pH approximately 6) has been identified: N-(β-D-ribofuranosyl)-formamide. Thin layer silica gel chromatography and spectroscopic methods (mass spectrometry, NMR, IR, optical rotation) enabled to isolate and characterize this product. It may be assumed that this substance resulted from the hydrolytic degradation of unstable hydroxy-hydroperoxides. It was also prepared by uridine oxydation with pertrifluoracetic acid. A similar major compound in the radiolysis of uridylic acid has been found

Abstract (French)

Le produit majeur d'irradiation gamma de l'uridine en solution aqueuse aeree (10-3M, pH approximativement 6) a ete identifie au N-(β-D-ribofurannosyl) formamide Ce compose proviendrait de la degradation hydrolytique d'intermediaires hydroxyhydroperoxydes instables selon un processus comparable a celui mis en jeu dans le cas de la thymidine. Le derive formamide a ete isole sur couche mince de silice. Il a ete caracterise par des methodes spectroscopiques (spectrometrie de masse, RMN, IR, pouvoir rotatoire). La synthese du produit a ete realisee par oxydation de l'uridine avec l'acide pertrifluoracetique prepare in situ. Un compose analogue le N-(phosphoryl-5-β-D-ribofurannosyl) formamide a ete trouve apres irradiation gamma de de l'acide uridylique
