Pharmacokinetic study and interaction with the reticulo-endothelial system of a plasma substitute: modified fluid gelatine
Description
By the labelling of a plasma substitute prepared from gelatine, MFG or modified fluid gelatine, it was possible to establish certain facts concerning the kinetics of MFG and its action on the cells of the reticulo-endothelial system. Iodine labelling proved sufficiently reliable to allow the pharmakokinetic exploration of the product by a method which, being relatively simple to use, offers good reproducibility of results. Various tests on the strength of the iodine protein bond showed the stability of the entire protein molecule. A study of the metabolism revealed that the MFG in question behaves towards the organism not as a pure substance but as a mixture of two molecular species of different mass. The experimental results concerning the influence of MFG on the reticulo-endothelial system showed that the phagocyte function is only modified if there is competition between the phagocyted molecule and the MFG

Le marquage d'un substitut du plasma, prepare a partir de gelatine, la G.F.M. ou gelatine fluide modifiee, a permis de mettre en evidence un certain nombre d'informations concernant la cinetique de la G.F.M. et son action sur les cellules du systeme reticulo endothelial. Le marquage a l'iode s'est revele etre une methode suffisamment fidele pour permettre l'exploration pharmacocinetique du produit par une technique dont la mise en oeuvre relativement simple permet une bonne reproductibilite des resultats. Les divers controles effectues sur la solidite de la liaison iode proteine ont permis de mettre en evidence la stabilite de la molecule proteique toute entiere. L'etude du metabolisme a montre que la G.F.M. etudiee ne se comportait pas vis-a-vis de l'organisme comme un produit pur mais comme le melange de deux especes moleculaires differant par leur masse. Les resultats experimentaux obtenus lors de l'influence de la G.F.M. sur le systeme reticulo endothelial ont montre que la fonction phagocytaire n'etait modifiee que lorsqu'il existait une competition entre la molecule phagocytee et la G.F.MFiles
