Mathematical model applied to decomposition rate of RIA radiotracers: 125I-insulin used as sample model

Description

A mathematical model is described to fit the decomposition rate of labelled RIA compounds. The model was formulated using four parameters: one parameter correlated with the radioactive decay constant; the chemical decomposition rate 'K*' of the radiolabelled molecules; the natural chemical decomposition rate 'K' and; the fraction 'f*' of the labelled molecules in the substrate. According to the particular values that these parameters can assume, ten cases were discussed. To determine one of these cases which fit the experimental data, three types of samples were need: radioactive; simulated radiotracer ('false radiolabelled') and; on labelled common substrate. The radioinsulin 125I was used as an example to illustrate the model application. The experimental data substantiate that the insulin labelled according to the substorchiometric procedures and kept at freezer temperature were degraded with K=0.45% per day. (Author)

Formulou-se um modelo matematico para descrever a cinetica da degradacao de tracadores moleculares radioativos utilizados em radioensaios. Para a formulacao deste modelo foram considerados quatro parametros: um parametro associado a constante de desintegracao do radionuclideo; a constante 'K*' de degradacao independente da radiodesintegracao; a constante 'K' da degradacao quimica da molecula na sua forma natural e; a fracao 'f*' na amostra das moleculas marcadas. Levando-se em conta os valores que estes quatro parametros podem assumir, foram analisados dez casos. Para enquadrar os dados experimentais a um dos dez casos determinou-se a concentracao em diferentes tempos da amostra radioativa, de uma amostra fria simuladora (proveniente de uma falsa marcacao) e de amostra fria controle. A fim de ilustrar a aplicacao deste modelo utilizou-se a radioinsulina marcada com 125I. Esta amostra degradou-se com taxa diaria de 0,45% e a fracao das moleculas marcadas com o 125I foi de 72%.
