Studies the thermal late effect and the mode of recovery of mild and severe hyperthermia with using nuclear and bio-molecular techniques in male rabbits

Hyperthermia plays a significant role in the evidence-based on treatment of cancer patients. The rationale for the use of hyperthermia as a treatment for cancer comes from in vitro and early clinical studies suggesting that tumour cells may be more sensitive to high temperature compared with normal cells. The prospect of using hyperthermia alone to treat cancer tumorous is appealing because hyperthermia is a physical treatment and so would have fewer side effects than chemotherapy or radiotherapy and, it could be used in combination with these therapeutic approaches.The main objective of this study was to investigate the thermal kinetics of mild and severe whole body hyperthermia (WBH) at 42 degree C and 43 degree C and its thermal late effect, at 7, 14, 21 and 28 days post mild and severe WBH (42 degree C and 43 degree C) in normal strain male rabbits, on some biochemical parameters, DNA and RNA in (liver + kidney) and heat shock proteins. Besides the histological structure of testis, liver kidney and heart before and after mild and severe WBH treatment and its thermal late effects at 7, 14, 21 and 28 days. It was attempted to evaluate the degree of safety and the effective time when hyperthermia is applied in the clinic for treating cancer and other diseases with keeping the normal cell intact. Normal 74 adult local strain male rabbits were used in this study. They were treated with mild whole body hyperthermia (WBH at 42 degree C) and severe WBH (43 degree C) induced by heat exposure (46 degree C± 1 degree C, 18-23 % RH) for about 1-1.5 hr or whichever reach first. The blood samples (1.5ml) were collected from each rabbit before and immediately after both mild (42 degree C) and severe (43 degree C) WBH treatments and at 7, 14, 21 and 28 days post heat exposure. Each rabbit served as its own control. The following blood serum parameters were estimated: alanine amino transferase (ALT); aspartate amino transferase (AST) lactate dehydrogenase (LDH), creatine phosphokinase (CPK) activities, total serum protein, albumin, globulin, blood urea nitrogen (BUN) and creatinine. Beside the estimation of DNA and RNA in the liver and kidney, as well as the testosterone hormone, body weight (BW) and rectal temperature (RT) were recorded for each period of time. The heat shock proteins (HSPs) were also determined in the serum.
