Effects of in vivo priming on in vitro induction of cytotoxicity, 2

Responding spleen cells of C3H/He mice presensitized with AKR tumor cells were cultured for 5 days together with mitomycin-C-treated stimulating cells of C57BL/6 origin. Cultured lymphocytes exhibited cytotoxicity not only to the presensitizing and in vitro stimulating cells but also to the third party cells of allogeneic or xenogeneic origin. Antigen-specificity of these cytotoxic lymphocytes was examined by the inhibition of destruction of 51Cr-labelled target cells with unlabelled cells. Unlabelled cells of the presensitizing and in vitro stimulating cell donors inhibited cytotoxicity not only to the presensitizing and in vitro stimulating cells but also to the third party cells. Cytotoxicity to a third party target was inhibited with the homologous third party cells but not with another third party cells. The possible explanations on such a phenomenon are discussed. (author)
