Cu(I) assisted nucleophilic exchange radiohalogenation: Application and mechanistic approach
Description
The basis of the Cu(I) assisted nucleophilic exchange is the presence of an excess of reducing agents in acidic conditions to produce in situ Cu(I) from CuSO4 and maintain Cu(I) during the entire reaction. This method avoids generation of higher oxidation states of iodine and of degradation products. It has been applied with success for 123I - Kits (IAMP, HIPDM, MIBG, Hippuran, PFFA) and for the production of 123I labeled brain receptor tracers. The mechanism is supposed to be a two step reaction: (i) complex formation and (ii) exchange, while the overall labeling reaction follows a pseudo first order. This method is also applied for the cold synthesis of 2-I-Ketanserin and 2'-I-spiperone
