Study of PIG3 expression and cell cycle redistribution in AHH-1 cells induced by neutron irradiation with different energy

Description
p53 is a well-known tumor suppressor gene. PIG3 is downstream gene of p53 induced by DNA damage and play important role in many physiological and pathological responses, such as cell cycle arrest, apoptosis, aging and so on. Previous studies suggested that PIG3 is a promising biological dosimeter. In present study, the PIG3 expression and cell cycle redistribution of AHH-1 irradiated by thermal or fast neutrons were investigated, to provide experimental basis for PIG3 as a new biological dosimeter applied in neutron dosimetry. The results showed that PIG3 expression induced by thermal or fast neutron increased in linear-quadratic correlation with dose increased, and the quantity or time of appearance for fast neutrons was much more and earlier than that of thermal neutrons. Both of them also induced G2/M arrest in similar time-response with each of PIG3 expression. Moreover, G1 arrest was also detected in fast neutron irradiated AHH-1 and followed G2/M arrest. In conclusion, the effect on AHH-1 from fast neutrons was more significant than that of thermal neutrons. (authors)
