Influence of substances offering protection against radiation-induced delayed damage to the liver of the mouse

The influence of various radiation protection substances, among them cystamine, WR 638, WR 2721 and polymer-bound WR 2721, on the formation of liver tumours was investigated on a histological basis in long-term experiments in male mice following wholebody irradiation at the 2.5 Gy dose level, in the 7 Gy to 8 Gy dose range and at the 15 Gy level as well as following irradiation of the liver region alone with a dose of 5 Gy. Tumours in the liver region were observed to develop no earlier than 170 days after exposure. With the exception of a dextrane (WR 2721) conjugate/amine, there were no indications whatsoever that radiation protection substances may prevent the occurrence of radiation-induced liver tumours or reduce the tumour rate. The available body of evidence appears to suggest that liver tumours largely are primary changes. The radiation-induced hepatic tumours found in the mice studied showed great histological resemblance to those caused in man by toxic substances or the influences of thorotrast. Abstract (German)
In Langzeitversuchen an maennlichen Maeusen wurde der Einfluss verschiedener Strahlenschutzsubstanzen, wie Cystamin, WR 638, WR 2721 und von polymergebundenem WR 2721, auf die Bildung von Tumoren in der Leber nach Ganzkoerperbestrahlung mit Dosen von 2,5 Gy, 7 bis 8 Gy sowie 15 Gy und nach Teilkoerperbestrahlung der Leberregion mit 5 Gy histologisch untersucht. Fruehestens 170 Tage nach der Bestrahlung entwickelten sich Tumoren im Leberbereich. Abgesehen von einem Dextran-(WR 2721)-Konjugat/Amin ergaben sich keinerlei Hinweise, dass die Strahlenschutzstoffe die Bildung von strahleninduzierten Lebertumoren verhindern oder die Tumorrate vermindern. Nach den vorliegenden Daten entwickeln sich die Lebertumoren im wesentlichen primaer. Die strahlenbedingten Lebertumoren der Maeuse entsprechen histologisch weitgehend den durch toxische Stoffe oder durch Thorotrasteinwirkung beim Menschen entstandenen Lebertumoren.
