Determination of catecholamines and their metabolites by radioisotopic techniques, value in pharmacology and physiopathology
Description
At present the only way to estimate catecholamines and similar compounds at concentrations between 10 and 100 femtomoles is by the use of radioenzymatic techniques. Such methods are all based in practice on the enzymatic transformation of these substrates, in the presence of labelled S-adenosylmethionine, under the action of catechol-O-methyltrans-ferase (COMT) or phenylethanol-amine-N-methyltransferase (PNMT), which means that molecules suitable for such determinations must possess either a catechol group (catecholamines, dihydroxyphenylalanine, dihydroxyphenylacetic acid) or a phenylethanolamine group (noradrenaline, methoxynoradrenaline). At present the largest number of molecules may be estimated by methods based on the principle of O-methylation by COMT. The main processes described in the literature are examined, with special reference to the proposed means of solving problems which arise at various stages of the determination, mention is made of the many difficulties inherent in this kind of manipulation and of the limits to be expected of these tests. The immunological aspect of quantitative research on catecholamines and their derivatives is mentioned, work in this direction at present being based on radioimmunology. As a practical illustration of the many methodological studies mentioned, the application of radioisotopic techniques to in vitro exploration of the catecholamine metabolism is discussed; the contribution of these new techniques is shown particularly in the physiopathological study of certain metabolic disorders observed in man, in the pharmacodynamic study of certain molecules and in experimental studies on the central nervous system

A l'heure actuelle, les techniques radioenzymatiques sont les seules capables de permettre l'appreciation des catecholamines et des composes voisins a des taux compris entre 10 et 100 femtomoles; elles sont toutes basees, en pratique, sur la transformation enzymatique de ces substrats, en presence de S-adenosylmethionine marquee, sous l'action de la catechol-O-methyltransferase (COMT) ou de la phenylethanol-amine-N-methyl-transferase (PNMT); c'est-a-dire que les molecules susceptibles d'etre determinees selon un tel procede devront presenter soit un groupement catechol (catecholamines, dihydroxyphenylalanine, acide dihydroxyphenylacetique), soit un groupement phenylethanolamine (noradrenaline, methoxy-noradrenaline). Les methodes actuelles qui permettent d'apprecier le plus grand nombre de molecules, sont basees sur le principe de l'O-methylation par la COMT. Les principaux procedes decrits dans la litterature sont etudies en considerant les solutions proposees pour resoudre les problemes poses au niveau des differentes etapes du dosage. Il est fait mention des nombreuses difficultes inherentes a ce type de manipulation et des limites qu'il faut savoir reconnaitre a ces essais. L'aspect immunologique de l'etude quantitative des catecholamines et de leurs derives est evoque, car les tentatives realisees dans ce sens sont actuellement basees sur la radio-immunologie. Afin de concretiser le resultat des multiples travaux methodologiques evoques, l'application des techniques radio-isotopiques a l'exploration in vitro du metabolisme des catecholamines est abordee; l'apport de ces techniques nouvelles est mis en evidence en particulier dans l'etude physiopathologique de certains troubles metaboliques observes chez l'homme, dans l'etude pharmacodynamique de certaines molecules et dans les etudes experimentales au niveau du systeme nerveux centralAdditional details
