Influence of Lymphopoiesis Control Mechanisms on the Recovery of Immune Capacities in Isogenic Radiation Chimeras

Description

The capacities of cells belonging either to foetal liver or to adult bone marrow or spleen are extremely different as far as the production of immunologically competent cells (ICC) by isogenic chimeras is concerned. The study of thymectomized radiation chimeras by the Simonsen test and the Jern technique shows that it is only the spleen or the lymph nodes which are able to bring ICC to a thymectomized animal; the bone marrow and the foetal liver carry only precursor cells which cannot be transformed into ICC when the recipient lacks its thymus. Furthermore, the functional ICC brought to these animals by the spleen cell suspensions have limited life span, not exceeding 300- 400 days. It must also be stressed that the thymectomized isogenic chimeras treated by bone marrow or foetal liver, although they display no immune reactivity, have an increased production of immunoglobulins. Therefore it seems that in this case there is no regulation of the production, differentiation and maturation of ICC. However, a control of the production and/or reactivity of ICC does exist in non-thymectomized radiation chimeras. This can be shown by the Simonsen test and the Jern technique for the primary reactions and by the study of homologous tumour graft rejection for the second-set reaction. This regulatory activity could be mediated either through a feed-back mechanism acting on ICC production or through a more complex process involving an interaction between antigens and cells. Abstract (French)

Les potentialités des cellules, d'une part, du foie foetal et, d'autre part, de la moelle osseuse ou de la rate adulte sont extrêmement différentes en ce qui concerne la production de cellules immunologiquement compétentes (CIC) par les radiochimères isogéniques. L'étude des radiochimères thymiprives par le test de Simonsen et la technique de Jern montre que seule la rate ou les ganglions sont susceptibles de fournir des CIC à un animal dépourvu de thymus; en revanche la moelle osseuse et le foie foetal apportent des éléments précurseurs qui ne peuvent devenir des CIC que si le receveur est porteur d'un thymus. En outre, chez ces animaux les CIC fonctionnelles apportées par la rate ont une durée de vie limitée qui n'excède pas 300 à 400 jours. Paradoxalement, les radiochimères isogéniques thymiprives qui sont restaurées par la moelle osseuse et le foie foetal et sont dépourvues de réactivité immunologique ont un taux d'immunoglobulines supérieur à la normale. Il semble donc que, dans ce cas, il ne peut y avoir de régulation dans la production, la cytodifférenciation et la maturation des CIC. En revanche, chez les radiochimères non thymiprives, il existe un contrôle de la production ou de la réactivité des CIC ou de l'une et de l'autre, qui peut être mis en évidence aussi bien par le test de Simonsen que par la technique de Jern, en ce qui concerne les réactions primaires, et par le rejet d'une tumeur homotransplantable, dans le cas des réactions anamnestiques. Cette régulation pourrait se faire soit par un mécanisme de « feed-back» portant sur la production des CIC, soit par un processus plus complexe dans lequel interviendraient les concentrations respectives en CIC et en antigène.
