Hepatic antioxidant status of rats subjected to gamma radiation And / Or Cadmium Chloride And The Possible Protective Effect Of The α-Hexapod

The present study aims to detect the possible protective role of oral administration of α-hepadox on the histopathological changes in liver and to restore the activity of hepatic antioxidant status of rats subjected to 2 Gy gamma radiation and / or subcutaneous injection of 5 or 20 μmol/kg cadmium chloride (CdCl2). The obtained results showed significant reduction in the glutathione (GSH) and superoxide dismutase (SOD) activity and elevation of malonaldehyde (MDA) level in liver tissue due to gamma irradiation (2 Gy) as compared to control rats all over the experimental period (1 and 3 days post-irradiation exposure). The subcutaneous injection of CdCl2 in normal rat showed significant increase in GSH level all over the experimental periods as well as increase in the level of SOD at the dose level of 5 μ mol/kg. At the dose level of 5 or 20μmol/kg CdCl2 , MDA was decreased significantly after 72 hours. The subcutaneous administration of CdCl2 at dose level of 5 or 20 μmol/kg, directly after exposure of rats to 2 Gy gamma radiation, induced significant increases in the level of GSH and SOD and decreased MDA level. However, a significant increase was observed 72 hours post using the level of 20 μmol/kg CdCl2 . Oral administration of a-hepadox for one week after treatment of rats either with gamma rays and/or CdCl2 showed noticeable amelioration in the radiation-induced changes in most of the biochemical studied parameters. The histopathological studies revealed non-significant changes in the liver of rats treated with CdCl2 . Vacuolation of hepatic cells and presence of pecnotic nuclei in response to radiation exposure were observed. Amelioration in hepatic des function, associated with drastic histopathological changes in rats due to treatment with both stressors, was ameliorated by the use of α- hepadox
