Study of the metabolism of oxoplatinum-191 Pt

  • 1. All-Union Oncologic Science Center, Academy of Medical Sciences of the USSR, Moscow

Description

The purpose of the present investigation was to study the metabolism of the drug oxoplatinum and to determine the mechanism of its action. solve this problem, the authors used preparations of DDP and oxoplatinum labeled with the isotope191 Pt. The results of the investigations show that a preparation of oxoplatinum undergoes complex biological conversions in the organism, forming at least four metabolites, one of which is DDP, which also exhibits antitumor activity. The absence of nephrotoxicity for oxoplatinum, despite the formation of DDP in vivo is evidently due to the formation of a DDP-oxoplatinum complex, which leads to more rapid excretion of DDP and products of its metabolism from the kidney tissue, without causing irreversible changes in their structure
