Fixation, retention and exhalation of carrier-free 11C-labelled carbon monoxide by man

Description

Carrier-free 11C-labelled carbon monoxide was produced by proton irradiation of a nitrogen gas flow target via the 14N(p,α)11C process followed by on-line reduction of the predominantly formed 11C-carbon dioxide with a yield of 0.4 mCi/μAmin. After appropriate quality control about 2 mCi of carrier-free 11C-carbon monoxide in 500 ml of nitrogen gas were inhaled by test subjects in one breath. The 11C-activity distribution was then followed in vivo by scanning above thorax, head, liver, thigh and os sacrum; simultaneously the 11C-activity of the blood was also followed by batch measurement. The data indicate that part of the 11C-activity migrates from the blood into the intercellular space, while another part is exhaled. The 11C-activity leaves the individual organs with a biological half-life ranging from about 120 to 200 min, a time which is short as compared to the one observed for 51Cr-labelled erythrocytes. A radio gas chromatographic analysis of the exhaled air showed that the 11C-activity leaves the body exclusively in the form of 11C-labelled carbon monoxide. Consequently, metabolism of the 11CO into 11Co2 or other compounds can be excluded. (orig.)

Abstract (German)

Traegerfreies 14C-markiertes Kohlenmonoxid wurde durch Protonenbestrahlung eines stroemenden Stickstoffgastargets ueber den Prozess 14N(p,α)11C und anschliessende On-line-Reduktion des dabei hauptsaechlich entstandenen 11C-Kohlendioxid mit einer Ausbeute von 0,4 mCi/μAmin hergestellt. Nach einer angemessenen Qualitaetskontrolle wurden etwa 2 mCi traegerfreies 11C-Kohlenmonoxid in 500 ml Stickstoffgas von Versuchspersonen in einem Atemzug eingeatmet. Dann wurde die 11C-Aktivitaetsverteilung in vivo durch Abtasten ueber Brust, Kopf, Leber, Huefte und Kreuzbein verfolgt; gleichzeitig wurde auch die 11C-Aktivitaet im Blut durch Batch-Messungen verfolgt. Die Daten zeigen, dass ein Teil der 11C-Aktivitaet vom Blut in den interzellulaeren Raum abwandert, waehrend ein anderer Teil ausgeatmet wird. Die 11C-Aktivitaet verlaesst die einzelnen Organe mit einer biologischen Halbwertzeit von etwa 120-200 Minuten, einer im Vergleich zu 51C-markierten Erythrozyten kurzen Zeitspanne. Eine radiogaschromatographische Analyse der ausgeatmeten Luft zeigte, dass die 14C-Aktivitaet den Koerper ausschliesslich in Form von 11C-markiertem Kohlenmonoxid verlaesst. Ein Metabolismus des 11CO zu 11CO2 oder anderen Verbindungen kann somit ausgeschlossen werden. (orig./AK)
