Development of a new radio-immunologic proof for fibrinopeptide A
Description
Because of the cross-reaction of antiserum with fibrinogen and other proteins, these proteins have to be separated from the plasma before the radio-immuno-assay (RIA) is carried out. Here a simplified and also much faster separation technique is introduced. A more efficient procedure is obtained, due to the RIA realization on micro titre plates. By separating the bound tracer from the free one by means of the double-antibody method, even more precise results could be achieved. In patients suffering from phlebothrombosis, infarction or hepatocirrhosis, the FpA concentrations were measured by applying the described simplified and rationalized RIA. The proposed RIA appears to be appropriate for routine examinations. (MG)

Wegen der Kreuzreaktion des Antiserums mit Fibrinogen und anderen Proteinen muss vor der Durchfuehrung des Radioimmunoassays (RIA) eine Trennung dieser Proteine vom Plasma erfolgen. Eine vereinfachte und zugleich schnellere Methode zur Abtrennung wird vorgestellt. Durch Ausfuehrung des RIA auf Mikrotiterplatten wird eine rationellere Arbeit ermoeglicht. Durch Trennung des gebundenen vom freien Tracer mittels Doppel-Antikoerper-Methode konnten noch genauere Ergebnisse erzielt werden. Mittels des beschriebenen vereinfachten und rationalisierten RIA wurden bei Phlebothrombose-, Herzinfarkt- und Leberzirrhosepatienten die FpA-Konzentrationen gemessen. Der vorgeschlagene RIA scheint fuer Routineuntersuchungen geeignet. (MG)Additional details
