Significance of poly(ADP-ribosylation) enzymes and their inhibitors on heavy ion therapy

Description

Deficiency of poly(ADP-ribose) polymerase-1 (Parp-1) and poly(ADP-ribose) glycohydrolase (Parg) in embryonic stem (ES) cells both show sensitizing effect on g-irradiation. The impact of PARP inhibitor and functional inhibition of Parp-1 and Parg on sensitivity to carbon-ion irradiation was studied. PARP inhibitor caused sensitization effect on carbon-irradiation in MIAPaCa2 pancreatic cancer cells. Parg and Parp-1 deficient ES cells also showed enhanced sensitivity to carbon-ion irradiation. These results suggest that PARP inhibitor and Parg functional inhibition will be useful for the sensitization to heavy ion irradiation. (author)
