Radiommunoassay for triiodothyronine in serum. Development of the solid phase technic and comparison with two liquid phase RIA systems: the polyethylene glycol (PEG) and double antibody methods

Description

A solid phase radioimmunoassay (RIA) system for triiodothyronine (T3) was established by immobilizing triiodothyronine antibodies on the inner wall of reaction tubes. The antibody-coated tubes were made via reaction of antibody with glutaraldeyde residue pre coated on the inner wall of the tubes by alkaline self-polimerization. The quality of the coated tubes was tested through its performance in RIA methodology, by analysing the following RIA parameters: minimum detectable dose (MMD), nonspecific binding (NSB), X 50%, slope of the standard curve, intra and inter assay precision, accuracy of the method and figure of merit. The quality and characteristics of the reagents used in the RIA were analysed. (M.A.C.)

Estabeleceu-se um sistema de radioimunoensaio de hormonio triiodotironina (T3) pela tecnica de 'Fase Solida', imobizando-se anticorpo anti-T3 em tubos de polipropileno pre recobertos com glutaraldeido polimerizado em meio alcalino. Avaliou-se a qualidade dos tubos recobertos pelo seu desempenho metodologico no radioimunoensaio (RIE), analisando-se: a dose minima detectavel, a ligacao inespecifica ('NSB'), 'X50%', inclinacao da curva ('Slope'), precisao intra e inter ensaio, exatidao do medoto e digito de merito. Avaliou-se a qualidade e as caracteristicas dos reagentes utilizadas nos RIES. (M.A.C.)
