Kinetics and mechanism of mono and diiodination of tyrosine by molecular iodine and chloramine-T/I-

Description

The observed rate constant ratio, K1obs/K2obs, for the mono- to diiodination of tyrosine has been determined by the use of 3H and 14C-tyrosine and product analysis by HPLC. The K1obs/K2obs values at pH 7.50 and 5.00 were in the range 1.31 ± 0.06 to 1.20 ± 0.04 for chloramine-T/I- iodinations and 1.17 ± 0.09 to ± 0.83 ± 0.22 for molecular iodine iodinations. Chloramine-T/I- iodinations in water/ethanol mixtures have shown an increase in K1obs/K2obs with increased ethanol content. This is in contrast to the contention that the preferential diiodination of buried tyrosyls in proteins is due to the decreased polarity of the microenvironment. 13 refs., 3 tabs., 2 figs
