Evaluation of Beta tubulin expression in cell lines (SiHa, CaSKi and HeLa) exposed to UV-B light

Description
The ultraviolet light when interacting with the cells of the organism produce free radicals, therefore the exhibition to high UV doses cause different damages to the organism, causing the cellular death, senescence among others. In this way the UV-B light is more harmful than the UV-A and with the passage of time they are perceiving higher doses of said light. The purpose of this study was to evaluate the expression of Beta tubulin in cell lines subjected to UV-B radiation fields at a time and dose range ranging from 0 seconds taking it as control, 8 s (3.84 E-3 J/cm2), 10 s (4.8 E-3 J/cm2), 12 s (5.76 E-3 J/cm2), 14 s (6.72 E-3 J/cm2) and 20 s (9.6 E-3 J/cm2). One subjected to radiation, they were incubated under standard conditions (5%, CO2 and 37 degrees Celsius) so that they could process the damage of said agent. After 48 hours after the cells had been irradiated, the quantification of proliferation was carried out in each of the doses and thus to evaluate the behavior before the stress agent. Once the quantification of cell proliferation was carried out, the cells were lysed to evaluate the expression of cellular cytoskeleton proteins such as beta tubulin (TUBB). TUBB is a 55 kDa protein and acts as a guide in the transport of substances from one place to another in the cell and in the displacement of the organelles around the cytoplasm when associated with motor proteins such as kinesin and dynein. The SDS-Page Western Blot protocol was performed for the detection of TUBB expression and in this way to understand the effect of radiation on the cells derived from the cervix. The behavior at the cellular and molecular level in the cell lines was different, observing different patterns of proliferation and expression of TUBB at the established doses. (Author)
