Development of multi-residue method for the determination of pesticides in cereal matrices by isotopic dilution associated to liquid chromatography coupled to tandem mass spectrometry after pressurized liquid extraction

Pesticides are nowadays considered as toxic for human health. The maximal residues levels authorized in water and foodstuff are more and more strict. Therefore, selective analytical techniques are necessary for their identification and their quantification. The aim of this thesis was to develop a multi-residue method for the determination of pesticides by isotopic dilution associated to liquid chromatography (LC) coupled with tandem mass spectrometry (MS2) after pressurized liquid extraction. In a first step, an analysis method of phenyl-ureas and triazines in groundwater by isotopic dilution associated to LC/MS2 after solid phase extraction has been developed. The method has been validated and evaluated by participating to an inter-laboratory study. Concerning cereals, an extraction method of pesticides associated to an analysis by LC/MS2 has been developed for thirty-eight pesticides representative of twenty-six chemicals families and fourteen labeled compounds in wheat. The analysis has been carried out in reversed phase chromatography. Separation and detection conditions have been optimized. A global analytical protocol consisting of a pressurized liquid extraction step using acetonitrile at 100 deg. C and at 100 bars and followed by purification step of the resulting extract on a polymeric sorbent was developed. The developed method enables to extract thirty-eight pesticides and fourteen labeled compounds from wheat with recovery yield about 85% (RSD =4%, n=3). Moreover, the results show that the application of isotopic dilution can be complex. Although some improvements need to be added regarding experimentation of aged matrices or contaminated samples, the potential of the method has been demonstrated. (author)

Les pesticides sont aujourd'hui reconnus comme ayant des effets nefastes sur la sante humaine. Les limites maximales de residus (LMR) de pesticides autorisees dans l'eau et les aliments sont de plus en plus faibles. Ainsi, des techniques analytiques tres performantes sont indispensables pour leur identification et leur quantification. Cette these avait pour objectif de developper une methode d'analyse multi-residus de pesticides dans les matrices cerealieres par chromatographie en phase liquide (CPL) couplee a la spectrometrie de masse en tandem (SM2) apres une etape d'extraction en solvant chaud pressurise. Une premiere methode d'analyse de phenylurees et de triazines dans les eaux souterraines par dilution isotopique (DI) associee a la CPL/SM2 apres une etape d'extraction sur phase solide a ete developpee. Celle-ci a ete validee par la participation a un essai inter-laboratoires. Une methode d'extraction de pesticides dans le ble associee a une analyse par CPL/SM2 a ensuite ete developpee pour trente-huit pesticides representatifs de vingt-six familles chimiques et quatorze de leurs molecules marquees. La chromatographie a polarite de phase inversee a ete mise en oeuvre. Les conditions de separation et de detection ont ete optimisees. Un protocole analytique global comprenant une etape d'extraction a l'acetonitrile a chaud (100 deg. C) sous pression (100 bars) suivie d'une etape de purification de l'extrait obtenu sur un adsorbant de nature polymerique a ete mise en oeuvre. Par cette approche, il a ete possible d'analyser l'ensemble des molecules ciblees avec un taux de recuperation moyen de l'ordre de 85% (CV = 4%, n=3). Si ces resultats sont tres satisfaisants, ils ont, par ailleurs, souligne que l'application de la dilution isotopique est complexe. Malgre, quelques ameliorations a apporter notamment concernant differents essais sur matrices vieillies ou contaminees, le potentiel de la methode developpee a ete demontre. (auteur)Files
