Antimicrobial activity of organic extracts isolated from Aplysina fistularis (Demospongiae: Aplysinidae)

Description
Organic extracts of the sponge Aplysina fistularis (Pallas 1766), were tested for antimicrobial activity against Gram positive bacteria (Staphylococcus aureus) and Gram negative bacteria (Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa). The minimal inhibitory concentration (MIC) and toxic activity of bioactive extract were determined. Susceptibility trials of organic fractions obtained by VLC. Hexane, EtOAc and CHCl3 showed that EtOAc fraction has antibacterial activity against E. coli, while CHCl3 fraction inhibited E. coli and S. aureus growth. The later refractioning of EtOAc fraction and the biodirected assays showed that fractions F12 and F13 of EtOAc/Hex and EtOAc F14 were bioactive against E. coli, K. pneumoniae and S. aureus. Only EtOAc/MeOH Sf2 from subfractioning of EtOAc F14 produced inhibition for E. coli and S. aureus. In Sf2 EtOAc/MeOH, MIC was moderate for S. aureus (MIC > 256 μg/ml). F4 CHCl3/MeOH produced a high inhibition in S. aureus (MIC = 0.125 μg/ml) and for E. coli (MIC > 16 μg/ml). F10 CHCl3/MeOH showed a moderate activity against S. aureus (MIC > 128 μg/ml) and low activity against E. coli (MIC = 512 μg/ml). F10 CHCI3/MeOH did no present toxic activity against Artemia salina. The fractiorts F4 CHCI3/MeOH and Sf2 EtOAc/MeOH were toxic for this organism when the concentration was higher than 100 μg/ml. LC50 in both cases was 548.4 and 243.4 μg/ml respectively. The conclusion is that secondary metabolites of medium polarity obtained from A. fistularis have a wide spectrum of anti bacterial activity. Toxicity analysis suggests that only F10 CHCI3/MeOH has potential as an antimicrobial agent for clinical use. (author)

Los extractos organicos de la esponja Aplysina fistularis (Pallas 1766), fueron estudiados en relacion con su actividad antimicrobiana frente a bacterias Gram positiva (Staphylococcus aureus) y Gram negativas (Escherichia coli, Klebsiella pneumoniae y Pseudomonas aeruginosa). La concentracion minima inhibitoria (CMI) y la actividad toxica de los extractos bioactivos fueron determinadas. La fraccion organica fue cromatografiada por CLV. Las pruebas de susceptibilidad de las fracciones obtenidas con EtOAc, CHCl3 y Hexano, mostraron que la fraccion EtOAc posee actividad antibacteriana frente a E. coli, mientras que la fraccion CHCl3 inhibio el crecimiento de E. coli y S. aureus. El ulterior fraccionamiento de esta fraccion y los bioensayos biodirigidos mostraron que las fracciones F12 y F13 de EtOAc/Hex y la F14 EtOAc fueron bioactivas frente a E. coli, K. pneumoniae y S. aureus. El mayor halo de inhibicion fue producido por la F14 EtOAc frente a E. coli y S. aureus. La Sf2 EtOAc/MeoH proveniente del subfraccionamiento de F14 EtOAc produjo amplios halos de inhibicion para E. coli y S. aureus. La CMI de Sf2 EtOAc/MeOH resulto moderada para S. aureus (CMI > 256 μg/ml). La F4 CHCl3MeOH provoco una alta inhibicion para S. aureus (CMI = 0.125 μg/ml) y para E. coli (CMI > 16 μg/ml). La F10 CHCl3/MeOH mostro una moderada actividad frente a S. aureus (CMI > 128 μg/ml) y baja actividad por E. coli (CMI = 512 μg/ml). La F10 CHCl3/MeOH no presento actividad toxica frente a A. salina. Las fracciones F4 CHCl3/MeOH y Sf2 EtOAc/MeOH resultaron toxicas para este organismo a una concentracion mayor de 100 μg/ml. La LC50 para ambos casos fue de 548.4 y de 243.40 μg/ml, respectivamente. La conclusion es que las fracciones de mediana polaridad obtenidos de A. fistularis poseen amplio espectro de actividad antibacteriana. El analisis de toxicidad refleja que solo la F10 CHCl3/MeOH posee un alto potencial como agente antimicrobiano de uso clinico. (autor)Additional details
