Pulse radiolysis study of the reduction mechanism of an antitumor antibiotic, mitomycin C

Description
Mitomycin C is a quinonic antitumor metabolized in vivo by one-electron reduction. We have studied the mechanism of the one-electron reduction of this drug by pulse radiolysis using C00.- free radicals as reductants. Semiquinonic and hydroquinonic intermediates are formed. The hydroquinonic form undergoes a methanol elimination leading to a transient which can disappear in one of two ways: by either internal redox reaction or hydrolysis of the aziridine. 17 refs

La mitomycine C est un antibiotique antitumoral possedant un cycle quinonique et metabolise in vivo par reduction supposee monoelectronique. Nous avons etudie le mecanisme de la reduction monoelectronique de ce medicament, par radiolyse pulsee, les reducteurs etant les anions radicalaires C00.-. Des intermediaires semiquinonique puis hydroquinonique, sont tout d'abord formes. La forme hydroquinonique subit ensuite une elimination de methanol menant a un compose transitoire qui evolue par deux voies paralleles: soit par oxydo-reduction interne, soit par hydrolyse du cycle aziridineAdditional details
