Analysis of FDG by HPLC/MS

Description
In this presentation are shown the analysis results of the model solutions of 2-deoxy-2-[18F]fluoro-D-glucose (FDG) and other saccharides, obtained by an HPLC/ESI-MS (Agilent 1100 HPLC with electrospay ionization mass spectrometer granted by the IAEA) equipment, in serial connection with other detectors (flow scintillation detector with PET cell, UV-VIS diode array detector). As eluent was used CH3CN:0,025 % NH4COOH aq. (80:20). Retention time of FDG was estimated on Shodex Asahipak NH2P-50 column at 5.5 min. There were analysed also some other saccharides, which could be theoretically present in the real solution (glucose, fructose, galactose, mannose). The samples were prepared in the mobile phase at concentration of 0.1 -1 mg/ml. After the HPLC separation, as the most intensive mass spectrum ions were found, for all measured sugars: (M + NH4+)+, (2M + Na+)+ in positive and (M + COOH-)-, (2M - H-)- in negative ionization mode. In some special cases, when the column was unable to chemically separate the epimers, the chromatographic peaks were overlapped and the identification of compounds was not possible with the use of other detectors (UV-VIS, or refractometric), the mass spectra allowed the identification of some saccharides, according to the ratio of these ion - associates.
