Development of cardiac troponin-I radioimmunoassay and preliminary results in clinical application

Description

A double antibody radioimmunoassay (RIA) was developed to measure cardiac troponin-I (CTnI) in serum. CTnI from fresh human myocardium was purified and identified. Antisera against CTnI were raised in NZ rabbits with affinity constant (Ka) 4.1 x 10-10 L/mol. Iodination of CTnI was carried out with iodogen. The specific activity of the product was 3659 GBq/g. The ED40 was 6.4 +- 0.34 μg/L (x-bar +- s, n = 8) from standard curves. The sensitivity was 1.5 μg/L and the assay covered the range 2∼64μg/L for CTnI. The intra-assay and inter-assay coefficient of variation (CV) were 2.1%∼5.7% (n = 8) and 5.4%∼9.9% (n = 4) respectively. The assay recovery was 90.1%∼95.3%. Non-immunochemical cross reactivity with skeletal troponin-I(STnI) was observed. The normal value was 2.17 +- 0.66 μg/L (x-bar +-s) in sera of 87 normal persons
