Quantitation and renewal of alveolar and bronchiolar cell populations of rat lungs. Changes during some pathological processes

Description

The various cells of alveolar and bronchiolar tissues of rat lungs were studied qualitatively and quantitatively. In physiological conditions, the renewal rate of the cell populations is low and the frequency of the various cell types is constant. This stability, especially at the level of the alveolar tissue, was also found during the latency period and the development of radiation-induced lung cancers. A particular cellular population was demonstrated: marginated leukocyte pool at the level of the pulmonary circulation. This pool was different both qualitatively and quantitatively from the leukocytes of the systemic circulation and, in physiological conditions, behaved as a cellular reservoir of monocytes chiefly re-distributed according to the body needs. In pathological conditions, its fast migration contributed to the defence of the alveolar medium. A quantitative study of the renewal of alveolar macrophages showed that under 1 p. cent of the marginated leukocyte pool is used daily to keep up this population. This fraction undergoes a maturation stage by cellular division within the endoalveolar medium. In some pathological conditions, this division can be completely inhibited

Abstract (French)

Cette etude caracterise et quantifie les differentes cellules qui constituent les tissus alveolaires et bronchiolaires du poumon de rat. Elle montre que dans les conditions physiologiques ces populations presentent un faible taux de renouvellement et que la frequence des differents types cellulaires observes est constante. Cette stabilite des populations cellulaires, notamment au niveau du tissu, alveolaire, est retrouvee pendant le temps de latence et durant le developpement de cancers pulmonaires radioinduits. Une population cellulaire particuliere a pu etre mise en evidence: un pool leucocytaire margine au niveau des capillaires de la petite circulation. Ce pool se distingue quantitativement et qualitativement des leucocytes du sang circulant et se comporte, dans les conditions physiologiques, comme une reserve cellulaire notamment de monocytes redistribues selon les besoins de l'organisme. Dans les conditions pathologiques, sa migration rapide assure la defense du milieu endoalveolaire. L'etude quantitative du renouvellement des macrophages alveolaires montre qu'en fait moins de 1% du pool des monocytes margines est utilise quotidiennement pour entretenir cette population. Cette fraction subit une etape de maturation par division cellulaire au sein du milieu endoalveolaire. Dans certaines conditions pathologiques, cette division peut etre totalement inhibee
