Characterization of uranium tolerant bacteria in TBP-Kerosene organic phase of uranium waste

Description

Characterization of uranium tolerant bacteria in TBP-Kerosene organic phase of uranium waste has been investigated. The purpose of this research was to characterize and identify uranium tolerant bacteria from uranium waste. The uranium tolerant bacteria was selectively isolated from enrichment medium which is consist of 10 % v/v uranium waste in liquid glucose media then ten colonies were isolated. Each colony was sub cultured in isolation and screening agar plate then the pure culture isolates of uranium tolerant bacteria was resulted. Each pure culture was grown on Nutrient Broth growth medium which contain of uranium in varies concentration i.l; 0 ppm,120 ppm, 150 ppm, and 180 ppm. The isolate with the highest instantaneous growth constant in concentration 180 ppm of uranium was the chosen of uranium tolerant bacteria and then was characterized. Characterization of bacteria isolates consist of colony morphology, cell morphology, gram staining and biochemical tests. The result of characterization was identified by profile matching method with references genera which are traced in Bergey's Manual of Determinative. The identification resulted that the isolates of uranium tolerant bacteria include in genera of Pseudomonas, Escherichia and Citrobacter. (author)
