Cellular radiation effects and hyperthermia: Cytokinetic investigations with stationary phase yeast cells

Description
Wild type diploid yeast, Saccharomyces cerevisiae strain 211, was subjected to 250 kV X-rays or 500C heat treatment for 30 min or to a combination of both. X-ray exposure took place either in air or in nitrogen. Cell number, percentage of budding cells and cell cycle progression was followed for up to 12 h post irradiation. The distribution of cell cycle stages was determined by flow cytofluorometry. All treatments cause a retardation of cell division rate. Hyperthermia leads mainly to a lengthening of G1, whereas X-rays arrest the cells reversibly in G2. The effect of the combined treatment appears to be merely additive. No selective action of hyperthermia on hypoxic cells was found. (orig.)
Abstract (German)
Der diploide Stamm 211 des Wildtyps von Saccharomyces cerevisiae wurde 30 min mit 250 kV Roentgenstrahlung oder einer Erwaermung auf 500C oder beidem behandelt. Die Roentgenbehandlung wurde entweder an der Luft oder in Stickstoff durchgefuehrt. Die Zellzahl, der Anteil der knospenden Zellen und der Verlauf des Zellsystems wurde bis zu 12 h nach der Bestrahlung verfolgt. Die Verteilung der einzelnen Stadien des Zellzyklus wurde mit Hilfe der Durchflusszytofluorometrie bestimmt. Alle 3 Arten der Behandlung verringern die Zellteilungsrate. Die Hyperthermie bewirkt lediglich eine Verlaengerung der G1-Phase, waehrend Roentgenstrahlung zu einem reversiblen Verharren in der G2-Phase fuehrt. Die kombinierte Behandlung scheint lediglich einen additiven Effekt zu haben. Eine selektive Wirkung der Hyperthermie auf hypoxische Zellen wurde nicht festgestellt. (orig.)Additional details
