Chapter 19. Blood and bone marrow. C. Blood platelet kinetics
Description
The blood platelet life span was measured by labelling the circulating population in vivo and in vitro. DF32P labelling in vivo: DFP is a specific inhibitor of acetyl-cholinesterase and hence in vivo labels blood platelets in the same way as the red cells and white cells which contain this enzyme. Sodium chromate 51Cr: this is the method used almost universally and the various stages were described. Several parameters were studied: the percentage of blood platelets in circulation, the aspect of the radioactivity decay curve, blood platelet production. Results obtained by the use of a medulla tracer, 75Se selenomethionine, were also reported. Finally the practical use of the blood platelet kinetics measurements were demonstrated
Abstract (French)
La duree de vie des plaquettes a ete mesuree par des methodes de marquage de la population circulante in vivo et in vitro. Marquage in vivo par DF32P. Le DFP est un inhibiteur specifique de l'acetyl-cholinesterase, il marque donc in vivo les plaquettes tout comme les globules rouges et les globules blancs qui contiennent cette enzyme. Marquage in vitro par le radiochromate de sodium 51Cr. C'est la methode quasi universellement utilisee. On en a deduit les differentes etapes. Plusieurs parametres ont ete etudies: le pourcentage de plaquettes circulantes, l'aspect de la courbe de decroissance de la radioactivite, la production des plaquettes. On a rapporte egalement les resultats obtenus par l'utilisation d'un marqueur medullaire, la selenomethionine 75Se. On a montre enfin l'utilisation pratique des epreuves de mesure de la cinetique plaquettaireAdditional details
