The role of tryptophan in the photochemical crosslinking of peptides and proteins

Description
Irradiation of wool and several other proteins in the presence of 14C-labelled tryptophan results in the covalent binding of radioactive material to the protein. This result is consistent with the claim that irradiation of proteins introduces crosslinks involving tryptophan residues. Radiolabelling techniques have been used to study the products of irradiation of the tripeptide, Ala-14C-Trp-Val. Some of these products are polymeric - further evidence for the involvement of tryptophan in crosslinking reactions. Most of the yellow material produced during irradiation in air is polymeric. Bei der Bestrahlung von Wolle und anderen Proteinen in Gegenwart von 14C-markiertem Tryptophan entsteht eine kavalente Bindung zwischen dem radioaktiven Material und dem Protein. Dieses Ergebnis bestaetigt die Behauptung, dass bei der Bestrahlung von Proteinen Querverbindungen mit Tryptophan-Rueckstaenden entstehen. Die Bestrahlungsprodukte des Tripeptids Ala-14C-Trp-Val wurden mit radioaktiven Markierungsmethoden untersucht. Einige dieser Produkte sind polymerisch - ein weiterer Beweis fuer die Rolle des Tryptophans in den Querverbindungsreaktionen. Der groesste Teil des bei der Bestrahlung an der Luft entstehenden gelben Materials ist polymerisch. (orig./AK)
