Bone marrow stromal cell transplantation
Description
Bone marrow is the seat of hematopoiesis. Proliferation and differentiation of hematopoietic progenitors are controlled by the medullary microenvironment which is constituted by stromal cells acting through growth factors, and adhesion molecules. The medullary stromal cells that have primary functions in the control of normal hematopoiesis could be used to locally produce growth factors that would allow the stimulation of hematopoiesis. The study presented in this report first aims to assess the ability of these cells to implant in the bone marrow, then to consider the use of stromal cells as vectors to produce local growth factors to stimulate hematopoiesis. The SCID and NOD-SCID mice are immuno-tolerant, the subcutaneous transplantation of human fetal bone fragments allows the maintenance of human hematopoiesis in vivo in its own microenvironment for several months (SCID or NOD-SCID-hu) mice. NOD-SCID-hu mice were used as hosts for the study of stromal cell transplantation. Three types of stromal cells were used in this study: - a human line (MRC-5) expressing the gene for β-galactosidase, - a murine stromal line (MSS) that was transfected to produce IL3-hu and cells derived from human stromal precursors that have been retrovirally transduced and express CD2m. These cells were injected directly into human grafts, or intravenously into NOD-SCID-hu mice. Genetically modified stromal cells have been implicated in human fetal bone grafts but also in the bone marrow, liver and spleen of mice. MSS cells produced detectable IL3-hu in mouse key serum and their implantation in human grafts was correlated with stimulation of human hematopoiesis. This study has also shown that human medullary stromal cells can be retrovirally transduced and can be transplanted. These cells could therefore be proposed as a vector for locally producing a molecule of therapeutic interest
Abstract (French)
La moelle osseuse est le siege de l'hematopoiese. La proliferation et la differenciation des progeniteurs hematopoietiques sont controlees par le micro environnement medullaire qui est constitue par les cellules stromales agissant par l'intermediaire de facteurs de croissance, et de molecules d'adherence. Les cellules stromales medullaires qui ont des fonctions primordiales dans le controle de l'hematopoiese normale pourraient etre utilisees pour produire localement des facteurs de croissance qui permettraient la stimulation de l'hematopoiese. L'etude presentee dans ce rapport a tout d'abord pour but d'evaluer la capacite de ces cellules a s'implanter dans la moelle osseuse, ensuite, d'envisager l'utilisation des cellules stromales comme vecteurs pour produire localement des facteurs de croissance pour stimuler l'hematopoiese. Les souris SCID et NOD-SCID sont immunotolerantes, la greffe en position sous-cutanee de fragments d'os foetaux humains permet le maintien de l'hematopoiese humaine in vivo dans son propre microenvironnement pendant plusieurs mois (souris SCID ou NOD-SCID-hu). Les souris NOD-SCID-hu ont ete utilise comme hote pour l'etude de la greffe des cellules stromales. Trois types de cellules stromales ont ete utilisees dans cette etude: -une lignee humaine (MRC-5) exprimant le gene de la β-galactosidase, -une lignee stromale medullaire murine (MSS) qui a ete transfectee pour produire l'IL3-hu et -les cellules derivees des precurseurs stromaux humains qui ont ete transduites par voie retrovirale et expriment le CD2m. Ces cellules ont ete injectees directement dans les greffons humains, ou par voie intraveineuse chez la souris NOD-SCID-hu. Les cellules stromales genetiquement modifiees se sont implantees dans les greffons d'os foetal humain mais aussi dans la moelle osseuse, le foie et la rate des souris. Les cellules MSS ont produit de l'IL3-hu detectable dans le serum cles souris et leur implantation dans les greffons humains a ete correlee avec la stimulation de l'hematopoiese humaine. Cette etude a aussi permis de montrer que les cellules stromales medullaires humaines pouvaient etre transduites par voie retrovirale et qu'elles pouvaient etre transplantees. Ces cellules pourraient donc etre proposees comme vecteur pour produire localement une molecule d'interet therapeutique. (auteur)Files
