Osteokalzinexpression and regulation in hematologic malignancies and in cultured cells

Description

Main issue of this work was to gain further insight into the association of haematopoiesis and osteopoiesis. A crucial cue for that is the fact that haematopoietic stem cells of haematopoietic diseases, which are characterised by c-KIT (CD117) expression, express the osteoblast marker osteocalcin. Thus, attention was focussed on the expression and regulation of osteocalcin, on one hand in blood and bone marrow samples of haematological diseases and on the other hand in leukaemic and osteosarcoma cell lines, i.e., by 1. investigating the expression of osteocalcin (OCN) splicing variants in haematological malignancies. We analysed bone marrow obtained from two patients with chronic myeloid leukaemia (CML), seven patients with other myeloproliferative diseases (MPD) and four patients with acute myeloid leukaemia (AML). RT-PCR analyses were performed in order to assess and quantify spliced (OCNs) and unspliced (OCNu) mRNA, the associated transcription factors (AML1 and AML3) as well as c-KIT, which is a marker for activated stem cells. Our data indicate that OCNs mRNA and OCN protein are expressed in c-KIT positive neoplastic stem cells in haematological malignancies. 2. It has been suggested that the tyrosine kinase inhibitor imatinib mesylate (IM), which has proven anti-proliferative effect, influences osteogenesis and bone turnover in treated patients. Thus, we aimed to quantify OCN mRNA, its splicing variants, the associated Runt-domain transcription factors AML1 and AML3, c-KIT and several metabolic genes to gain evidence about the differentiation state in the HL-60 leukaemia cell line as well as MG63 and U2OS osteosarcoma cells and murine primary osteoblasts MC3T3-E1. Our data indicate that IM induces inhibition of proliferation and synthesis of total OCN-mRNA in all cell lines, but a relative increase of OCNs-mRNA was observed in the human cell lines. On the other hand, differentiation-associated genes appeared to be stimulated. This may also indicate an osteoprotective effect of IM in vivo, but a complex equilibrium has to be considered. Thus, several leukaemic cell-types and osteoblasts with a malignant potential such as osteosarcoma cells are influenced by consequences of IM-induced tyrosine kinase inhibition. (author)

Abstract (German)

Hauptziel der vorliegenden Arbeit war es, die gemeinsame Basis der Haematopoese und Osteopoese zu charakterisieren. Da haematopoetische Stammzellen haematologischer Erkrankungen, charakterisiert durch die Expression von c-KIT (CD117), auch den Osteoblastenmarker Osteokalzin exprimieren, lag das Hauptaugenmerk auf der Expression Osteokalzins und den dieser zugrundeliegenden Regulationsmechanismen. Diese sollten sowohl in Blut- und Knochenmarkszellen bei haematologischen Erkrankungen als auch in Leukaemie- und Osteosarkom-Zellinien untersucht werden, naemlich mittels: 1. quantitativer Bestimmung der mRNA-Expression des Osteoblastenmarkers Osteokalzin (OCN) sowie dessen Spleissvarianten in haematologischen Erkrankungen in Abhaengigkeit einerseits von der Art der Krankheit, andererseits vom Krankheitsstadium. Wir untersuchten Knochenmarksproben zweier Patienten mit chronisch myeloischer Leukaemie (CML), sieben mit anderen myeloproliferativen Erkrankungen (MPD) und vier mit akuter myeloischer Leukaemie (AML). Mit Hilfe von RT-PCR-Analysen sollten die Expressionsmuster der gespleissten (OCNs) und ungespleissten OCN-mRNA (OCNu), der assoziierten Transkriptionsfaktoren AML1 und AML3 (= sogenannte Runt-domain Faktoren) sowie des c-KIT (CD117), eines Markers fuer aktivierte Stammzellen, quantifiziert werden. Unsere Daten weisen darauf hin, dass OCNs und OCN-Protein in c-KIT-positiven neoplastischen Stammzellen von Patienten mit haematologischen Erkrankungen exprimiert werden. 2. Es wird vermutet, dass der antiproliferativ wirkende Tyrosinkinasehemmer Imatinibmesylat (IM) Erneuerung und Umbau der Knochen behandelter Patienten beeinflusst. Wir untersuchten die Auswirkungen von IM in vitro in der leukaemischen Zellinie HL60 und in den Osteosarkomzellen MG63 und U2OS, bzw. primaeren Maus-Osteoblastenzellen MC3T3-E1. Besonderes Augenmerk galt der Genexpression von Osteokalzin und dessen Spleissvarianten und der Transkriptionsfaktoren AML1 und AML3 sowie von c-KIT und einigen Stoffwechsel-assoziierten Genen, um Informationen ueber den Differenzierungsstatus der Zellen zu erhalten. Unserer Versuchsreihe zufolge wurden durch IM sowohl Zellproliferation als auch OCN-Expression in allen Zellinien gehemmt, hingegen wurde ein relativer Anstieg der OCNs-mRNA in den humanen Zellinien beobachtet. Gene, die eher in differenzierten Zellen exprimiert werden, wurden hingegen stimuliert. Damit laesst sich erklaeren, dass das Wachstum leukaemischer Zellen und Osteoblasten mit malignem Charakter von IM gehemmt, das Gedeihen gesunder Zellen der Haematopoese und Osteogenese hingegen gefoerdert wird, was auf eine osteoprotektive Wirkung von IM hinweisen koennte.
