32P dan 3H isotopic labeling to support the diagnosis of diseases caused by microorganisms infection

Description

An accurate diagnosis is an important tool in the effective control of infectious diseases such as tuberculoses (TB) and malaria, both of which are caused by microorganisms infection that lead to high number of deaths. In case of TB the resistance of bacteria to the drugs may have caused the harder management of the patients, whereas in case of malaria the microscopic misdiagnosis may have led to fatal consequences. Due to these limitations, it is important that a reliable and rapid method for diagnosis is developed such as isotopic labeling with 32P and 3H. In our research 15 samples of sputum of TB suspected patients and 20 P. falciparum infected blood of malaria were tested. Research showed that the 32P labeling of DNA of M. tuberculosis in single strand conformational polymorphism (SSCP) method to assess the resistance based on the mobility shift of DNA band on acrylamide gel helped to obtain relatively more accurate diagnosis. On other hand 3H labeled hypoxanthine incorporation to parasites in in vitro culture in the determination of percentage of erythrocyte infection post irradiation using low scintillation counter (LSC) method also have helped to obtain more accurate diagnosis. We conclude that even though has some limitations the labeling method has a special role in the uphold of diagnosis. (author)
