Light stress study on in vitro photoautotrophic cultures. Application to the search of mutants

Light stress applied on a photoautotrophic ce11 suspension of Euphorbia characias leads to inhibition of photosynthesis. The examination of the effects of O2 concentration and methyl viologen showed a limited implication of the superoxide radical. Moreover, photoinhibition occurs at non saturating light suggesting a major contribution of the donor side of the photosystem II. In long term, the energetic deficit induced by the prolonged photosynthesis photoinhibition results in a catabolic pathway that leads, in the absence of tissue cooperation, to the death of the cells. This work was used to establish on Solanum tuberosum a strategy for the in vitro selection of variants modified in their response to light stress. Photoautotrophic protocols were developed for the culture and regeneration of callus derived from protoplast as we11 as for the culture of a regenerating cell suspension. A light stress screening procedure under 2% CO2 was applied on the photosynthetically active callus derived from these two systems. In the regenerated plants, several physiological variants have been obtained, some of them showing different levels of photoinhibition under air or under 2% CO2 when compared to the wild type; most of the regenerated plants do not exhibit the higher photoinhibition which is observed, under 2% CO2, in the wild type. The effects of high CO2 and light in the screening procedure are discussed. (author)

Abstract (French)

Un stress lumineux applique sur des cellules d'une suspension photoautotrophe d'Euphorbia characias se traduit par une inhibition de la photosynthese. L'etude de l'effet de la concentration en O2 et du methyl viologen indique que le radical superoxyde serait peu implique. De plus, cette photoinhibition apparait en lumiere non saturante ce qui suggere une importante contribution du cote donneur du photosysteme II. A long terme, le deficit energetique engendre par l'inhibition de la photosynthese resulte en un catabolisme cellulaire qui se manifeste, en l'absence d'interaction tissulaire, par la mort rapide des cellules. Ces travaux ont ete utilises pour elaborer une strategie de selection in vitro de variants de Solanum tuberosum modifies dans leur reponse au stress lumineux. Des protocoles de culture en condition photoautotrophe ont ete mis au point sur un systeme protoplaste-cal-plante regeneree ainsi que sur une suspension cellulaire regenerante. Une procedure de criblage par application d'un stress lumineux sous 2% de CO2 a ete definie et mise en oeuvre sur les microcals photosynthetiques issus de ces differents systemes. Parmi les regenerants obtenus, il a ete.mis en evidence des variants physiologiques dont certains manifestent un niveau de photoinhibition different du sauvage sous air ou sous 2% de CO2; la plus forte photoinhibition observee chez le sauvage sous 2% de CO2 n'existe pas chez une majorite des regenerants. Les effets du fort CO2 et de la lumiere dans la procedure de criblage sont discutes. (auteur)
