Study On In Vitro Variation Generation Of Paphiopedilum By Ionization

Description

The lethal doses of 50% in vitro samples (LD50) of Paphiopedilum delenatii and Paphiopedilum callosum including protocorm like bodies (PLBs), shoot buds and plantlets were determined. The results for generation of in vitro variations of two mentioned orchids by irradiating PLBs, shoot buds and plantlets indicated that PLBs were the most suitable for this method and the treatment by ion beam (C6+) was much better than of by γ-ray from Co-60 source. Twelve in vitro variated lines of P. delenatii (with a frequency ca. 3.0 x 10-2) and 10 variation lines of P. callosum (with a frequency ca. 2.5 x 10-2) were obtained by irradiation of PLBs using ion beam (C6+) at a dose of 3 Gy.
