Description
Integrins, a family of transmembrane proteins have been gaining wide interest in radiobiological studies owing to their ability to facilitate cell adhesion, migration, and invasion. Recent studies have reported the importance of integrins in radiation response, making them potential targets for improving the efficacy of radiotherapy. In this review, we provide a robust understanding of the role of integrins in radiotherapy and its various molecular applications. In addition, we emphasize the mechanisms through which integrins mediate the signal transduction cascade events. Also, we review the role of integrins in the tumor microenvironment and how they contribute to the radioresistance of cancer cells. In addition, we review the strategies targeting integrins in combination with radiotherapy to overcome the resistance of cancer cells to ionizing radiation. Overall, this review summarizes the essence of integrins in radiotherapy and further suggests the challenges and future directions in this field. (author)

細胞膜貫通型タンパク質であるインテグリンは、細胞の接着・運動・浸潤等の機能に関与し、放射線生物学研究において注目され、興味深い知見が蓄積されつつある。近年、細胞の放射線応答や放射線耐性におけるインテグリンの重要性が次々と報告されてきており、放射線治療効果を向上させるための標的の一つとしても期待されている。本稿では、がん細胞の放射線治療抵抗性におけるインテグリンの役割と下流で活性化するシグナルの分子メカニズムについて概説する。さらに、がんの放射線治療に対する抵抗性の克服に向けて、インテグリンを標的とする治療戦略と将来の展望を考察する。(著者)Additional details
