Iodine quantification by analytical ion microscopy (AIM)

Description
AIM allows imaging and quantification of all elements on a biological tissue section. However quantification is limited by physical parameters which are difficult to assess and which intervene for a given element in the relation between its concentration and the measured signal. Thus, to quantify iodine in thyroid follicles, an iodine standard was prepared. Homogeneity in its atomic iodine distribution was tested by surface and depth analyses. Then, a standard curve was generated to be used as a reference for quantification of iodine on histological preparation by AIM
Abstract (French)
La MIA permet l'imagerie et la quantification de tous les elements de la classification periodique, presents dans une coupe de tissu biologique. Cependant, la quantification se heurte a la difficulte d'evaluer avec precision certains des parametres physiques intervenant, pour un element donne, dans la relation entre sa concentration atomique et l'intensite du signal mesure. Afin de mesurer la quantite absolue d'iode dans les follicules thyroidiens, un temoin iode a ete concu et evalue. L'homogeneite, dans la repartition atomique de l'iode, a ete verifiee. Puis, a partir de differentes dilutions, une courbe d'etalonnage a ete tracee afin d'etre utilisee comme reference pour la quantification de l'iode en MIAAdditional details
