Analysis of radiation-induced chromosomal aberrations in human lymphocytes after G-band staining

Human lymphocytes from a male donor were exposed in vitro to two different doses (1 Gy and 2 Gy) of hard (150 kV) X-rays in the G0 phase. The resulting chromosomal aberrations were evaluated in a total of 335 cells prepared by G-band staining. A total of 616 aberrations was found. As in earlier publications, there was a 2:1 ratio between interchromosomal and intrachromosomal aberrations. The fraction of complete interchromosomal processes amounted to 90%, and the ratio of associated acentric fragments to dicentric types was 0.90:1. The interchromosomal exchange aberrations have Poisson distributions as in earlier publications, and the relative frequencies of translocations, dicentric and extra-acentric chromosomes are compatible with a linear-quadratic dose-effect curve. Menschliche Lymphozyten eines maennlichen Spenders wurden in vitro in der G0-Phase harten (150 kV) Roentgenstrahlen zwei unterschiedlicher Dosierungen (1 Gy und 2 Gy) ausgesetzt. Die daraus resultierenden Chromosomenaberrationen wurden an insgesamt 335 mit der G-Banden-Faerbetechnik praeparierten Zellen detailliert ausgewertet, wobei zusammen 616 Aberrationsfiguren zu erkennen waren. In Uebereinstimmung mit frueheren Angaben verschiedener Autoren betrug das Verhaeltnis der inter- zu den intrachromosomalen Aberrationen ca. 2 : 1, der Anteil der komplett verlaufenen interchromosomalen Vorgaenge 90%, das Verhaeltnis assoziierter Azentrischer Fragmente zu dizentrischen Formen 0,90 : 1. Die Verteilung der interchromosomalen Austauschaberrationen folgen wie bei diesen Autoren Poissonverteilungen, und die relativen Haeufigkeiten der Translokationen, der dizentrischen und der extraazentrischen Chromosomen erscheinen kompatibel mit einer linear-quadratischen Dosis-Effekt-Beziehung.
