A new solid phase method for the determination of free thyroxine in serum using monoclonal antibodies (SPAC ET/FT4)

Description

A new assay for the determination of free thyroxine has been described. By using monoclonal antibodies with clearly defined properties and labelled thyroxine this assay allows fast and simple estimation of the hormone. Results of several studies indicated that the assay is able to clearly distinguish euthyroidism from hyper- and hypothyroidism with a tendency to underestimate very high hyperthyroid values. Sera of patients with anomalous binding proteins, such as analbuminemia, familial dysalbuminemic hyperthyroxinemia or increased/decreased TBG are classified correctly as euthyroid. The same is true for sera of patients suffering from non thyroid illness (NTI). Comparison to dialysis of the new assay system shows good correlation, especially in those cases with anomalous binding protein properties. (orig.)

Abstract (German)

Es wird ein neuer Test beschrieben, der - unter Verwendung monoklonaler Antikoerper mit genau definierten Eigenschaften und des markierten Hormons selbst - die Bestimmung des freien T4 erlaubt. Der Test ist schnell und sehr einfach durchzufuehren. Die Ergebnisse mehrerer Untersuchungen zeigten, dass der Test sehr gut differenziert zwischen Euthyreosen, Hypothyreosen und Hyperthyreosen, mit einer geringen Tendenz, hoeher hyperthyreote Werte etwas zu unterschaetzen. Seren von Patienten mit Proteinanomalien, wie Analbuminaemie, familiaere dysalbuminaemische Hyperthyroxinaemie oder TGB-Erniedrigung bzw. -Erhoehung, werden von diesem Assay richtig gemessen, ebenso Seren von Patienten mit nicht-schilddruesenspezifischen Erkrankungen. Der Vergleich dieses neuen Assaysystems mit der Dialyse zeigt auch gerade in den Faellen mit anormalen Proteinbindungsverhaeltnissen eine gute Korrelation. (orig.)
