Description

The recent discovery of substances able to compete with opiates for morphine-specific binding sites in the brain has opened up a new path for studying the made of action of these receptors. These morphinomimetic substances known as endorphines might be the precursors of smaller molecules, e.g. encephalines, two pentapeptides (H-Tyr-Gly-Gly-Phe-Met-OH or Met-Enc and H-Tyr-Gly-Gly-Phe-Leu-OH or Leu-Enk) which serve as neurotransmitters of the analyesic effect. A radioimmunoassay has been developed with the aim of measuring the distribution of these substances in the brain and of following their development in different states of psychopathology. (AJ)

Abstract (German)

Mit Hilfe eines verbesserten empfindlichen und spezifischen RIA fuer Encephaline unter Verwendung von jodierten Tracern kann die Konzentration dieser Substanzen in verschiedenen Teilen des Rattenhirns gemessen werden. Neuere Arbeiten, in denen Endorphine mit Hilfe des Radiorezeptorverfahrens gemessen werden, zeigen eine analoge Verteilung von Encephalinen, allerdings mit hoeheren Konzentrationen. Es scheint, dass mit diesem Verfahren auch andere Reaktionsprodukte mit einer Affinitaet fuer die Encephalin-Rezeptoren gemessen werden, wie z.B. α- und β-Endorphine. Die Weiterentwicklung eines Tests fuer diese Substanzen koennte diese Hypothese bestaetigen und unsere Kenntnisse ueber die Wirkung der Endorphine erweitern. (AJ)
