Effects of ionizing radiation on expression of γ-H2AX in jurkat cell lines

Description

Objective: To investigate the effects of ionizing radiation on the expression of γ-H2AX in Jurkat cell lines. Methods: The changes of the expressions of γ-H2AX in Jurkat cell lines after X-irradiation with different doses(0.5, 1.0, 2.0, 4.0, and 6.0 Gy) and after irradiation with 2.0Gy X-rays for different time (0, 0.5, 1.0, 4.0, 8.0, 16.0, and 24h) were analyzed by flow cytometry(FCM), and the apoptotic rate of Jurkat cell lines was detected during dose-effect experiment. Results: In time-effect experiment, the results showed that the expression of γ-H2AX reached to the peak within 1h, followed by a time-dependent decrease, kept expressing at 16.0h, and had no significant difference with control at 24.0h. In dose-effect experiment, it was demonstrated that the expressions of γ-H2AX showed a dose-dependent increase in the range of 0.5-4.0 Gy at 1.0 hafter exposed to X-rays, and dropped at the does of 6.0 Gy; at 8.0 hafter exposure to those doses above, the express levels of γ-H2AX enhanced linearly with the dose increased in the range of 0-4.0 Gy, showing a clear dose-dependent manner (P<0.01). Conclusion: The γ-H2AX expression could be increased by X-irradiation in time-dependent and dose-dependent manners. (authors)
