The effect of phosphonoformic acid on wild type and mutant S49 cell lines
Description
In contrast to the results with mammalan DNA polymerases in cell free systems, mammalian cell DNA synthesis is very resistant to PFA inhibition, requiring two orders of magnitude higher concentrations of PFA to achieve 50% inhibition. The authors studied the mechanism of inhibition of cellular replication by PFA by selecting mutant cell lines resistant to growth inhibition by phosphonoformic acid, and compared them with wild type S49 cells that are more sensitive to PFA inhibition. Ribonucleoside diphosphate reductase activity was measured by the conversion of CDP to deoxy CDP and 0.42 uC1 C-14 CDP
