High field nuclear magnetic resonance application to polysaccharide chemistry
Description
Nuclear magnetic resonance has been applied to polysaccharide chemistry using time averaging technique and high fields (100 and 250 MHz). The three methyl signals of methyl cellulose and cellulose triacetate are separated, and the C-6 substituent has been identified. Biosynthesis of bacterial cellulose has been performed using deuterium labelled D-glucose and Acetobacter xylinum. Per-acetylated derivative of bacterial cellulose has been studied by NMR; this study permitted us to determine the quantity of deuterium on each position of the anhydro-glucose unit in the polymer. NMR has also been used to see the anomeric end chain of cellulose and amylose derivatives and to show the fixation of bromine and t-butyl group on the free anomeric end chain of cellulose triacetate. Abstract (French)
Ce travail a consiste, tout d'abord, dans le but d'ameliorer la sensibilite de la RMN dans le cas des polysaccharides a employer la methode d'accumulation des spectres dans un analyseur multicanaux, puis la resonance a hauts champs (100 MHz et 250 MHz), pour l'etude des derives de la cellulose: acetates et triacetates. La RMN a permis de differencier les trois substituants des positions 2, 3 et 6 de l'unite glucose dans le cas ou les substituants sont des groupes methyles (methylcellulose) ou acetate (triacetate de cellulose). Dans une troisieme partie, l'utilisation de D-glucose marque au deuterium, dans la biosynthese de la cellulose par l'Acetobacter xylinum a permis de mettre en evidence sur le derive triacetyle les positions d'incorporation des atomes de deuterium sur le cycle glucose. Enfin, la RMN a ete utilisee dans l'etude des extremites de chaines et de la reactivite de ces extremites pour verifier que l'extremite anomere des polysaccharides presente: d'une part, une reactivite specifique; d'autre part que le proton porte par le carbone anomere a un deplacement chimique specifique. La RMN permet de suivre la fixation d'un atome de brome puis d'un groupe tertiobutyle a l'extremite anomere libre du triacetate de cellulose. (auteur)Files
