Significance of poly (ADP-ribosylation) enzymes and their inhibitors on heavy ion therapy

Description
Poly(ADP-ribose) polymerase-1 (Parp-1) deficient embryonic stem (ES) cells and Poly(ADP-ribose) glycohydrolase (Parg) deficient ES cells both show increased sensitivity to gamma-irradiation. We investigated the impact of Parp-1 and Parg functional inhibition on responses to heavy ion irradiation. Parp-1 knockout mouse embryonic fibroblast (MEF) exhibited increased levels of gamma-H2AX foci, a DNA double strand break marker, compared to wild-type MEF. Parg knockout ES cells showed enhanced apoptosis after carbon-ion irradiation. These results suggest that Parp-1 and Parg could be the factors which determine the sensitivity to heavy ion irradiation. (author)
