The radioimmunoassay of oxytocin in human plasma
Description
A detailed description is given of a technique for the radioimmunoassay of oxytocin in unextracted human plasma. Synthetic oxytocin was first 125I-iodinated and then purified by column chromatography. Antibodies to oxytocin were produced in rabbits. Oxytocinase activity in late pregnancy plasma was inhibited by the addition of o-phenantrolin. The method could detect 10μU oxytocin/ml. No assayable hormone was detected in plasma samples from men or non-pregnant women. Oxytocin levels ranging from 15 to 100 μU/ml could be detected in plasma samples taken during pregnancy and labour.
