Labelling of oleic acid and triolein with radioiodine-131

Description

Labelling of triolein and oleic acid by addition of iodine mono chloride the olefinic bond has been tried in organic media. The mechanism of halogenation of alkenes by elemental halogens has been found to occur either via radical mechanism or via electrophilic addition of the halogen to the double bond, which needs to be thermally initiated. The parameters affecting the labelling yield investigated.It was found that the yield of labelling of triolein and oleic acid in methyl ethyl ketone at reflux by elemental iodine is about 50% after 2 hours. However, half an hour was found to be convenient as a heating time to avoid serious decomposition of the labelled product. The removal of unreacted radioiodine has been tried using dowex 1 x 8 in the chloride from. The labelled compound was mixed with 2 ml 25% human serum albumin and passed on the column.
