5-fluorouracil metabolism monitored in vivo by 19F NMR

Description

The use of 19F NMR to monitor the in vivo metabolism of the chemotherapeutic drug, 5-fluorouracil (5FU), in tumours and in the liver of mice is described. The results clarified a number of steps in the pathway of 5FU metabolism in tumour tissue and liver which provided useful information for elucidating the antitumour mechanism of action. Thus this 19F NMR technique could be extended to clinical medicine, providing a direct, noninvasive measurement of the metabolism of the fluorinated drug both at its site of action and at its site(s) of detoxification.
