the relation between hyperthermia and heat shock proteins in carcinoma cell lines with consideration of hyperthermia late effects on rabbits

Description

these investigations were designed to study the thermal kinetics of whole body hyperthermia (42.5 deg C) and its late effects after 0,1,2,4,8,24 and 48 hrs in local strain female rabbits . the following parameters were estimated for each rabbit before and after WBH; serum ALT,AST,ALP, LDH and CPK activities, serum levels of total protein , urea, and glucose, T3.T4 and cortisol, beside the heat shock proteins (HSPs). rectal temperature and body weight were recorded before and after WBH ,histological structure of heart was studied also in control rabbits and at 24 and 48 hours post hyperthermia. furthermore, in the present investigation the induction of heat shock proteins in cancer cell before and after heat exposure were studied using MCF7 breast cancer cell line and hepatocellular carcinoma cell line HePG2. these cells were exposed to heat at 42.5 deg c and 41.5 deg c for half an hour and 1 hour respectively.cells were collected at the end of exposure and returned to the medium in the incubator. heat shock proteins, especially 70 KD family, were detected using western blot technique at 0,1,2,4,6,8, and 24 h after heating . control cells were collected excluding step
