Degradation of 14C-parathion 'in vitro' by microorganisms isolated from a gley humic soil

It was determined 'in vitro' the degradation of Parathion by a bacterium and a fungus isolated from a sample of Gley Humic soil previously treated with repeated applications of the insecticide. In a qualitative colorimetric assay hydrolisis of parathion to p-nitrophenol just the bacterium gave a positive answer. In quantitative assays of 14C-parathion degradation in culture media containing both microorganisms, organic solvents extractions resulted in organic and aqueous phases, which were analysed by liquid scintillation counting and thin-layer chromatography. In a mineral salts medium plus buffer, the bacterium and the fungus behaved differently from the control, because part of the 14C-insecticide was metabolized to, at least, one metabolite and besides, the microorganisms presented smaller percentages of total recovery. The largest percentage of the radio carbon recovery from the extracts of the medium containing the fungus plus extract of yeast, was obtained from the aqueous phase and the existence of other metabolite was demonstrated by chromatograms of the organic phase. Foi determinada in vitro a degradacao do paration por uma bacteria e um fungo isolados de amostra de solo Gley Humico, previamente tratado com aplicacoes repetidas do inseticida. Em teste colorimetrico qualitativo de hidrolise do paration a p-nitrofenol, apenas a bacteria respondeu positivamente. Em ensaios quantitativos de degradacao do 14C-paration em meios de cultura contendo os dois microrganismos, extracoes com solventes organicos resultavam em uma fase organica e uma fase aquosa, que foram analisadas por contagem de cintilacao em liquido e por cromatografia em camada delgada. Em meio de sais minerais e tampao, a bacteria e o fungo comportaram-se diferentemente do controle por metabolizar parte do 14C-inseticida a pelo menos um metabolito e por apresentar sempre menor recuperacao total. A maior porcentagem de recuperacao do radiocarbono dos extratos do meio contendo fungo acrescido de extrato de levedura foi obtida a partir da fase aquosa, sendo que os cromatogramas da fase organica demostraram a existencia de outro metabolito.
