Changes of fluoresceindiacetat-hydrolysis and -transport in mammalian cells after ultra-violet- and gamma irradiation

Changes in the permeability of cell membranes and in the enzymatic acitvity of intact cells and their homogenates after irradiation were studied. CHO-cells, erythrocytes and their homogenates were irradiated with cobalt-gamma radiation and UV-light. It was found that the esterases are more radiosensitive in intact cells than in the corresponding homogenates. The decrease of the enzymatic activity after irradiation can be described by a one-hit curve, whereas the loss of transport activity can be resolved into two one-hit curves. The slope associated with the more sensitive target mainly determines the course of the composed transport curve. Mechanisms for the decreases in the rate constants are discussed. (orig.)
Abstract (German)
Es wurden die Veraenderungen in der Permeabilitaet von Zellmembranen und in der Enzymaktivitaet intakter Zellen und ihrer Homogenate nach Bestrahlung untersucht. CHO-Zellen, Erythrozyten und ihre Homogenate wurden mit Co-Gammastrahlung und UV-Licht behandelt. Dabei zeigte sich, dass die Esterasen in intakten Zellen strahlenempfindlicher sind als in den entsprechenden Homogenaten. Die Abnahme der Enzymaktivitaet nach Bestrahlung kann durch eine Eintrefferkurve beschrieben werden, der Verlust an Transportaktivitaet dagegen durch eine Zweitrefferkurve. Der Abfall beim empfindlicheren Target bestimmt im wesentlichen den Verlauf der gemeinsamen Transportkurve. Die moeglichen Mechanismen fuer die Abnahme der Geschwindigkeitskonstanten werden diskutiert. (orig.)Additional details
